Brain Clearance, Sleep, and Circadian Rhythms: Implications for Brain Health

Abstract

This chapter provides an overview on the implications of brain clearance, sleep, and circadian rhythms for the brain. It highlights brain clearance as a significant function of sleep, referencing how brain clearance becomes less efficient with aging, hypertension, and several pathological conditions. The chapter then discusses the anatomy of the brain clearance system, particularly cerebrospinal fluid, aquaporin-4 perivascular spaces, and the glymphatic system. It then elaborates on the enhanced brain clearance during sleep in relation to proposed drivers of brain clearance, such as synchronized vasomotion and norepinephrine. It lastly explains the relevance of brain clearance for brain disorders, such as neurodegeneration and Alzheimer’s disease.
